    Robust Onboard Visual SLAM for Autonomous MAVs

    Abstract. This paper presents a visual simultaneous localization and mapping (SLAM) system consisting of a robust visual odometry and an efficient back-end with loop closure detection and pose-graph optimization. Robustness of the visual odometry is achieved by utilizing dual cameras pointing different directions with no overlap in their respective fields of view mounted on an micro aerial vehicle (MAV). The theory behind this dual-camera visual odometry can be easily ex-tended to applications with multiple cameras. The back-end of the SLAM system maintains a keyframe-based global map, which is used for loop closure detec-tion. An adaptive-window pose-graph optimization method is proposed to refine keyframe poses of the global map and thus correct pose drift that is inherent in the visual odometry. The position of each map point is then refined implicitly due to its relative representation to its source keyframe. We demonstrate the efficiency of the proposed visual SLAM algorithm for applications onboard MAVs in ex-periments with both autonomous and manual flights. The pose tracking results are compared with the ground truth data provided by an external tracking system.

    Primeval very low-mass stars and brown dwarfs - IV. New L subdwarfs, Gaia astrometry, population properties, and a blue brown dwarf binary

    30 pages, 23 figuresWe present 27 new L subdwarfs and classify five of them as esdL and 22 as sdL. Our L subdwarf candidates were selected with the UKIRT Infrared Deep Sky Survey and Sloan Digital Sky Survey. Spectroscopic follow-up was carried out primarily with the OSIRIS spectrograph on the Gran Telescopio Canarias. Some of these new objects were followed up with the X-shooter instrument on the Very Large Telescope. We studied the photometric properties of the population of known L subdwarfs using colour-spectral type diagrams and colour-colour diagrams, by comparison with L dwarfs and main-sequence stars, and identified new colour spaces for L subdwarf selection/study in current and future surveys. We further discussed the brown dwarf transition-zone and the observational stellar/substellar boundary. We found that about one-third of 66 known L subdwarfs are substellar objects, with two-thirds being very low-mass stars. We also present the Hertzsprung-Russell diagrams, spectral type-absolute magnitude corrections, and tangential velocities of 20 known L subdwarfs observed by the Gaia astrometry satellite. One of our L subdwarf candidates, ULAS J233227.03+123452.0, is a mildly metal-poor spectroscopic binary brown dwarf: a ~L6p dwarf and a ~T4p dwarf.     Gene Therapy Corrects Mitochondrial Dysfunction in Hematopoietic Progenitor Cells and Fibroblasts from Coq9R239X Mice

Please note that the results of this manuscript have been submitted to patent protection (application number P201630630; title: “Uses of Coenzyme Q biosynthetic proteins”; date:05/16/2016).Recent clinical trials have shown that in vivo and ex vivo gene therapy strategies can be an option for the treatment of several neurological disorders. Both strategies require efficient and safe vectors to 1) deliver the therapeutic gene directly into the CNS or 2) to genetically modify stem cells that will be used as Trojan horses for the systemic delivery of the therapeutic protein. A group of target diseases for these therapeutic strategies are mitochondrial encephalopathies due to mutations in nuclear DNA genes. In this study, we have developed a lentiviral vector (CCoq9WP) able to overexpress Coq9 mRNA and COQ9 protein in mouse embryonic fibroblasts (MEFs) and hematopoietic progenitor cells (HPCs) from Coq9R239X mice, an animal model of mitochondrial encephalopathy due to primary Coenzyme Q (CoQ) deficiency. Ectopic over-expression of Coq9 in both cell types restored the CoQ biosynthetic pathway and mitochondrial function, improving the fitness of the transduced cells.     Oxidative discolouration in whole-head and cut lettuce: biochemical and environmental influences on a complex phenotype and potential breeding strategies to improve shelf-life

    Lettuce discolouration is a key post-harvest trait. The major enzyme controlling oxidative discolouration has long been considered to be polyphenol oxidase (PPO) however, levels of PPO and subsequent development of discolouration symptoms have not always correlated. The predominance of a latent state of the enzyme in plant tissues combined with substrate activation and contemporaneous suicide inactivation mechanisms are considered as potential explanations for this phenomenon. Leaf tissue physical properties have been associated with subsequent discolouration and these may be influenced by variation in nutrient availability, especially excess nitrogen and head maturity at harvest. Mild calcium and irrigation stress has also been associated with a reduction in subsequent discolouration, although excess irrigation has been linked to increased discolouration potentially through leaf physical properties. These environmental factors, including high temperature and UV light intensities, often have impacts on levels of phenolic compounds linking the environmental responses to the biochemistry of the PPO pathway. Breeding strategies targeting the PALand PPOpathway biochemistry and environmental response genes are discussed as a more cost-effective method of mitigating oxidative discolouration then either modified atmosphere packaging or post-harvest treatments, although current understanding of the biochemistry means that such programs are likely to be limited in nature and it is likely that they will need to be deployed alongside other methods for the foreseeable future

    Co-occurrence of mutations in NF1 and other susceptibility genes in pheochromocytoma and paraganglioma

    IntroductionThe percentage of patients diagnosed with pheochromocytoma and paraganglioma (altogether PPGL) carrying known germline mutations in one of the over fifteen susceptibility genes identified to date has dramatically increased during the last two decades, accounting for up to 35-40% of PPGL patients. Moreover, the application of NGS to the diagnosis of PPGL detects unexpected co-occurrences of pathogenic allelic variants in different susceptibility genes.MethodsHerein we uncover several cases with dual mutations in NF1 and other PPGL genes by targeted sequencing. We studied the molecular characteristics of the tumours with co-occurrent mutations, using omic tools to gain insight into the role of these events in tumour development.ResultsAmongst 23 patients carrying germline NF1 mutations, targeted sequencing revealed additional pathogenic germline variants in DLST (n=1) and MDH2 (n=2), and two somatic mutations in H3-3A and PRKAR1A. Three additional patients, with somatic mutations in NF1 were found carrying germline pathogenic mutations in SDHB or DLST, and a somatic truncating mutation in ATRX. Two of the cases with dual germline mutations showed multiple pheochromocytomas or extra-adrenal paragangliomas - an extremely rare clinical finding in NF1 patients. Transcriptional and methylation profiling and metabolite assessment showed an “intermediate signature” to suggest that both variants had a pathological role in tumour development.DiscussionIn conclusion, mutations affecting genes involved in different pathways (pseudohypoxic and receptor tyrosine kinase signalling) co-occurring in the same patient could provide a selective advantage for the development of PPGL, and explain the variable expressivity and incomplete penetrance observed in some patients

    Evaluation of appendicitis risk prediction models in adults with suspected appendicitis

    Background Appendicitis is the most common general surgical emergency worldwide, but its diagnosis remains challenging. The aim of this study was to determine whether existing risk prediction models can reliably identify patients presenting to hospital in the UK with acute right iliac fossa (RIF) pain who are at low risk of appendicitis. Methods A systematic search was completed to identify all existing appendicitis risk prediction models. Models were validated using UK data from an international prospective cohort study that captured consecutive patients aged 16–45 years presenting to hospital with acute RIF in March to June 2017. The main outcome was best achievable model specificity (proportion of patients who did not have appendicitis correctly classified as low risk) whilst maintaining a failure rate below 5 per cent (proportion of patients identified as low risk who actually had appendicitis). Results Some 5345 patients across 154 UK hospitals were identified, of which two‐thirds (3613 of 5345, 67·6 per cent) were women. Women were more than twice as likely to undergo surgery with removal of a histologically normal appendix (272 of 964, 28·2 per cent) than men (120 of 993, 12·1 per cent) (relative risk 2·33, 95 per cent c.i. 1·92 to 2·84; P < 0·001). Of 15 validated risk prediction models, the Adult Appendicitis Score performed best (cut‐off score 8 or less, specificity 63·1 per cent, failure rate 3·7 per cent). The Appendicitis Inflammatory Response Score performed best for men (cut‐off score 2 or less, specificity 24·7 per cent, failure rate 2·4 per cent). Conclusion Women in the UK had a disproportionate risk of admission without surgical intervention and had high rates of normal appendicectomy.     RelocNet: Continous metric learning relocalisation using neural nets

    We propose a method of learning suitable convolutional representations for camera pose retrieval based on nearest neighbour matching and continuous metric learning-based feature descriptors. We introduce information from camera frusta overlaps between pairs of images to optimise our feature embedding network. Thus, the final camera pose descriptor differences represent camera pose changes. In addition, we build a pose regressor that is trained with a geometric loss to infer finer relative poses between a query and nearest neighbour images. Experiments show that our method is able to generalise in a meaningful way, and outperforms related methods across several experiments
